Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.405A>C (p.Lys135Asn), citing Ambry Variant Classification Scheme 2023: The p.K135N variant (also known as c.405A>C), located in coding exon 5 of the BRCA1 gene, results from an A to C substitution at nucleotide position 405. The lysine at codon 135 is replaced by asparagine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
