Uncertain significance — the classification assigned by Ambry Genetics to NM_006648.4(WNK2):c.5742G>T (p.Gln1914His), citing Ambry Variant Classification Scheme 2023: The p.Q1914H variant (also known as c.5742G>T), located in coding exon 23 of the WNK2 gene, results from a G to T substitution at nucleotide position 5742. The glutamine at codon 1914 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
