Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000551.4(VHL):c.311G>A (p.Gly104Asp), citing Ambry Variant Classification Scheme 2023: The p.G104D variant (also known as c.311G>A), located in coding exon 1 of the VHL gene, results from a G to A substitution at nucleotide position 311. The glycine at codon 104 is replaced by aspartic acid, an amino acid with similar properties. This variant was determined to be functionally neutral in one saturation genome editing assay (Buckley M et al. Nat Genet, 2024 Jul;56:1446-1455). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
