Submitter rationale: This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 167 of the EARS2 protein (p.Cys167Tyr). This variant is present in population databases (rs397514594, gnomAD 0.008%). This missense change has been observed in individuals with leukoencephalopathy with thalamus and brainstem involvement and high lactate (PMID: 22492562). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 39792).
