Uncertain significance for Yunis-Varon syndrome — the classification assigned by Care4Rare-SOLVE, CHEO to NM_018052.5(VAC14):c.1895C>T (p.Thr632Met): These two variants were found in a compound heterozygous state. VAC14 forms tri-molecular complex with FIG4 and PIKFYVE. Mouse model shows vacuolar neurodegeneration. Biallelic missense cause striatonigral degeneration. Compelling candidate given CADD score, rarity, mouse model, and forms part of a complex with FIG4, a gene responsible for a portion of children with Yunis-Varon syndrome
