Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004807.3(HS6ST1):c.916C>T (p.Arg306Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 306 of the HS6ST1 protein (p.Arg306Trp). This variant is present in population databases (rs780352591, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with HS6ST1-related conditions. ClinVar contains an entry for this variant (Variation ID: 39690). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt HS6ST1 protein function with a negative predictive value of 80%. Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on HS6ST1 function (PMID: 21700882). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
