Uncertain significance — the classification assigned by Ambry Genetics to NM_001127208.3(TET2):c.3010A>G (p.Lys1004Glu), citing Ambry Variant Classification Scheme 2023: The p.K1004E variant (also known as c.3010A>G), located in coding exon 1 of the TET2 gene, results from an A to G substitution at nucleotide position 3010. The lysine at codon 1004 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
