Uncertain significance — the classification assigned by Ambry Genetics to NM_001127208.3(TET2):c.3015G>C (p.Lys1005Asn), citing Ambry Variant Classification Scheme 2023: The p.K1005N variant (also known as c.3015G>C), located in coding exon 1 of the TET2 gene, results from a G to C substitution at nucleotide position 3015. The lysine at codon 1005 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
