Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.158A>C (p.Gln53Pro), citing Ambry Variant Classification Scheme 2023: The p.Q53P variant (also known as c.158A>C), located in coding exon 1 of the TERT gene, results from an A to C substitution at nucleotide position 158. The glutamine at codon 53 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
