NM_198253.3(TERT):c.463C>T (p.Arg155Cys) was classified as Uncertain significance for Dyskeratosis congenita by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R155C variant (also known as c.463C>T), located in coding exon 2 of the TERT gene, results from a C to T substitution at nucleotide position 463. The arginine at codon 155 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was reported in a study of patients with sporadic idiopathic pulmonary fibrosis (Dressen A et al. Lancet Respir Med, 2018 Aug;6:603-614). This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
