Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.8153G>A (p.Gly2718Asp), citing Ambry Variant Classification Scheme 2023: The p.G2718D variant (also known as c.8153G>A), located in coding exon 55 of the ATM gene, results from a G to A substitution at nucleotide position 8153. The glycine at codon 2718 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
