This variant lies in the ODAD1 gene (transcript NM_001364171.2) at the canonical splice acceptor site of the intron immediately before coding-DNA position 598, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Disruption of this splice site has been observed in individual(s) with clinical features of primary ciliary dyskinesia (PMID: 23261302; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. ClinVar contains an entry for this variant (Variation ID: 39639). This variant is present in population databases (rs606231239, gnomAD 0.004%). This sequence change affects an acceptor splice site in intron 5 of the CCDC114 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in CCDC114 are known to be pathogenic (PMID: 23261302, 23261303).
