Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000455.5(STK11):c.657C>G (p.Phe219Leu), citing Ambry Variant Classification Scheme 2023: The p.F219L variant (also known as c.657C>G), located in coding exon 5 of the STK11 gene, results from a C to G substitution at nucleotide position 657. The phenylalanine at codon 219 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
