Uncertain significance — the classification assigned by Ambry Genetics to NM_006947.4(SRP72):c.586A>G (p.Met196Val), citing Ambry Variant Classification Scheme 2023: The p.M196V variant (also known as c.586A>G), located in coding exon 5 of the SRP72 gene, results from an A to G substitution at nucleotide position 586. The methionine at codon 196 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
