This variant lies in the CRPPA gene (transcript NM_001101426.4) at coding-DNA position 713, where C is replaced by T; at the protein level this means replaces threonine at residue 238 with isoleucine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
