Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005902.4(SMAD3):c.511G>C (p.Glu171Gln), citing Ambry Variant Classification Scheme 2023: The p.E171Q variant (also known as c.511G>C), located in coding exon 3 of the SMAD3 gene, results from a G to C substitution at nucleotide position 511. The glutamic acid at codon 171 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
