Uncertain significance — the classification assigned by Ambry Genetics to NM_001377540.1(SLMAP):c.2407A>G (p.Lys803Glu), citing Ambry Variant Classification Scheme 2023: The p.K769E variant (also known as c.2305A>G), located in coding exon 20 of the SLMAP gene, results from an A to G substitution at nucleotide position 2305. The lysine at codon 769 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
