Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.538A>G (p.Thr180Ala), citing Ambry Variant Classification Scheme 2023: The p.T180A variant (also known as c.538A>G), located in coding exon 1 of the SKI gene, results from an A to G substitution at nucleotide position 538. The threonine at codon 180 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
