Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.719C>T (p.Pro240Leu), citing Ambry Variant Classification Scheme 2023: The p.P240L variant (also known as c.719C>T), located in coding exon 1 of the SKI gene, results from a C to T substitution at nucleotide position 719. The proline at codon 240 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
