Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_152703.5(SAMD9L):c.161A>G (p.Asp54Gly), citing Ambry Variant Classification Scheme 2023: The p.D54G variant (also known as c.161A>G), located in coding exon 1 of the SAMD9L gene, results from an A to G substitution at nucleotide position 161. The aspartic acid at codon 54 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
