NM_001283009.2(RTEL1):c.242T>C (p.Phe81Ser) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F81S variant (also known as c.242T>C), located in coding exon 2 of the RTEL1 gene, results from a T to C substitution at nucleotide position 242. The phenylalanine at codon 81 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
