Uncertain significance — the classification assigned by Ambry Genetics to NM_001023.4(RPS20):c.43G>T (p.Val15Leu), citing Ambry Variant Classification Scheme 2023: The p.V15L variant (also known as c.43G>T), located in coding exon 2 of the RPS20 gene, results from a G to T substitution at nucleotide position 43. The valine at codon 15 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
