Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000321.3(RB1):c.166G>T (p.Asp56Tyr), citing Ambry Variant Classification Scheme 2023: The p.D56Y variant (also known as c.166G>T), located in coding exon 2 of the RB1 gene, results from a G to T substitution at nucleotide position 166. The aspartic acid at codon 56 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
