Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002878.4(RAD51D):c.455C>T (p.Ala152Val), citing Ambry Variant Classification Scheme 2023: The p.A152V variant (also known as c.455C>T), located in coding exon 5 of the RAD51D gene, results from a C to T substitution at nucleotide position 455. The alanine at codon 152 is replaced by valine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
