Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.1215A>T (p.Glu405Asp), citing Ambry Variant Classification Scheme 2023: The p.E405D variant (also known as c.1215A>T), located in coding exon 8 of the RAD50 gene, results from an A to T substitution at nucleotide position 1215. The glutamic acid at codon 405 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
