Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000314.8(PTEN):c.99T>G (p.Ile33Met), citing Ambry Variant Classification Scheme 2023: The p.I33M variant (also known as c.99T>G), located in coding exon 2 of the PTEN gene, results from a T to G substitution at nucleotide position 99. The isoleucine at codon 33 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
