Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.974G>A (p.Gly325Asp), citing Ambry Variant Classification Scheme 2023: The p.G325D variant (also known as c.974G>A), located in coding exon 7 of the PTCH1 gene, results from a G to A substitution at nucleotide position 974. The glycine at codon 325 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
