Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.3854A>T (p.Gln1285Leu), citing Ambry Variant Classification Scheme 2023: The p.Q1285L variant (also known as c.3854A>T), located in coding exon 31 of the POLE gene, results from an A to T substitution at nucleotide position 3854. The glutamine at codon 1285 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
