NM_006231.4(POLE):c.4193A>T (p.Tyr1398Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1398F variant (also known as c.4193A>T), located in coding exon 33 of the POLE gene, results from an A to T substitution at nucleotide position 4193. The tyrosine at codon 1398 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
