NM_006231.4(POLE):c.6310T>A (p.Phe2104Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F2104I variant (also known as c.6310T>A), located in coding exon 45 of the POLE gene, results from a T to A substitution at nucleotide position 6310. The phenylalanine at codon 2104 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
