Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.2398C>A (p.Pro800Thr), citing Ambry Variant Classification Scheme 2023: The p.P800T variant (also known as c.2398C>A), located in coding exon 14 of the PMS2 gene, results from a C to A substitution at nucleotide position 2398. The proline at codon 800 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
