NM_000384.3(APOB):c.659C>A (p.Thr220Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T220K variant (also known as c.659C>A), located in coding exon 6 of the APOB gene, results from a C to A substitution at nucleotide position 659. The threonine at codon 220 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
