Uncertain significance — the classification assigned by GeneDx to NM_000256.3(MYBPC3):c.3726G>T (p.Lys1242Asn), citing GeneDx Variant Classification (06012015). This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 3726, where G is replaced by T; at the protein level this means replaces lysine at residue 1242 with asparagine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the MYBPC3 gene. The K1242N variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The K1242N variant occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. Nevertheless, this substitution is a semi-conservative amino acid substitution, which may or may not impact secondary protein structure as these residues differ in some properties.
