Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000039.3(APOA1):c.400G>C (p.Glu134Gln), citing Ambry Variant Classification Scheme 2023: The p.E134Q variant (also known as c.400G>C), located in coding exon 3 of the APOA1 gene, results from a G to C substitution at nucleotide position 400. The glutamic acid at codon 134 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
