NM_005477.3(HCN4):c.322G>C (p.Gly108Arg) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the HCN4 gene. The G108R variant has not been published as pathogenic or been reported as benign to our knowledge. No data are available from control populations to assess the frequency of this variant in the general population. The G108R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. Although this substitution occurs at a position that is conserved in mammals, arginine (R) is the native amino acid at this position in several species. Furthermore, the majority of in silico models predict this variant likely does not alter the protein structure/function.
