Uncertain significance — the classification assigned by GeneDx to NM_001110556.2(FLNA):c.425G>T (p.Trp142Leu), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the FLNA gene. The W142L variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The W142L variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. However, to our knowledge no studies have been performed to determine the functional effect of the W142L variant.
