Likely pathogenic — the classification assigned by GeneDx to NM_001492.6(GDF1):c.371G>T (p.Cys124Phe), citing GeneDx Variant Classification (06012015): The C124F variant in the GDF1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The C124F variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In addition, this substitution occurs at a position that is conserved across mammalian species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. Adequate data is not available in large population cohorts to assess the frequency of this variant in publicly available databases; however, this variant has not been detected at a significant frequency in presumably healthy individuals tested at GeneDx. We interpretC124F as a likely pathogenic ariant.
