Uncertain significance — the classification assigned by GeneDx to NM_015215.4(CAMTA1):c.893G>A (p.Gly298Glu), citing GeneDx Variant Classification (06012015). This variant lies in the CAMTA1 gene (transcript NM_015215.4) at coding-DNA position 893, where G is replaced by A; at the protein level this means replaces glycine at residue 298 with glutamic acid — a missense variant. Submitter rationale: The G298E variant in the CAMTA1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G298E variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The G298E variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is not conserved and in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. Therefore, we interpret G298E as a variant of uncertain significance.
