NM_001166108.2(PALLD):c.1965-12957A>G was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q25R variant (also known as c.74A>G), located in coding exon 1 of the PALLD gene, results from an A to G substitution at nucleotide position 74. The glutamine at codon 25 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
