NM_001166108.2(PALLD):c.2515G>A (p.Asp839Asn) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D335N variant (also known as c.1003G>A), located in coding exon 5 of the PALLD gene, results from a G to A substitution at nucleotide position 1003. The aspartic acid at codon 335 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
