Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.17063C>G (p.Ala5688Gly), citing Ambry Variant Classification Scheme 2023: The p.A4731G variant (also known as c.14192C>G), located in coding exon 54 of the OBSCN gene, results from a C to G substitution at nucleotide position 14192. The alanine at codon 4731 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
