Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.397G>A (p.Val133Met), citing Ambry Variant Classification Scheme 2023: The p.V133M variant (also known as c.397G>A), located in coding exon 1 of the OBSCN gene, results from a G to A substitution at nucleotide position 397. The valine at codon 133 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
