Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.3641T>G (p.Met1214Arg), citing Ambry Variant Classification Scheme 2023: The p.M1122R variant (also known as c.3365T>G), located in coding exon 10 of the OBSCN gene, results from a T to G substitution at nucleotide position 3365. The methionine at codon 1122 is replaced by arginine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
