This variant lies in the SPRED1 gene (transcript NM_152594.3) at coding-DNA position 281, where T is replaced by C; at the protein level this means replaces isoleucine at residue 94 with threonine — a missense variant. Submitter rationale: The I94T variant in the SPRED1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The I94T variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The I94T variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Isoleucine are tolerated across species.
