Pathogenic — the classification assigned by GeneDx to NM_020699.4(GATAD2B):c.1045C>T (p.Gln349Ter), citing GeneDx Variant Classification (06012015). This variant lies in the GATAD2B gene (transcript NM_020699.4) at coding-DNA position 1045, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 349 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q349X variant in the GATAD2B gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q349X variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret Q349X as a pathogenic variant.
