NM_002253.4(KDR):c.2266+1G>T was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the KDR gene (transcript NM_002253.4) at the canonical splice donor site of the intron immediately after coding-DNA position 2266, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.2266+1G>T variant in the KDR gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This splice site variant destroys the canonical splice donor site in intron 15. This splice site variant is predicted to cause abnormal gene splicing resulting in an in-frame protein product with an abnormal message. However, in the absence of RNA/functional studies, the actual effect of c.2266+1G>T in this individual is unknown. The c.2266+1G>T variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.2266+1G>T as a variant of uncertain significance.
