Uncertain significance for Autosomal dominant Parkinson disease 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198578.4(LRRK2):c.4448G>A (p.Arg1483Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with glutamine at codon 1483 of the LRRK2 protein (p.Arg1483Gln). The arginine residue is weakly conserved and there is a small physicochemical difference between arginine and glutamine. This variant is present in population databases (rs113431708, ExAC 0.001%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. ClinVar contains an entry for this variant (Variation ID: 39190). This missense change has been observed in individual(s) with Parkinson disease (PMID: 18197194).
