This variant lies in the TGFB3 gene (transcript NM_003239.5) at coding-DNA position 11, where A is replaced by G; at the protein level this means replaces histidine at residue 4 with arginine — a missense variant. Submitter rationale: Variant summary: TGFB3 c.11A>G (p.His4Arg) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 250834 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.11A>G in individuals affected with Loeys-Dietz Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Three ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
