Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_016042.4(EXOSC3):c.239T>A (p.Val80Asp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: EXOSC3 c.239T>A (p.Val80Asp) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 8.5e-06 in 235192 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.239T>A in individuals affected with Pontocerebellar Hypoplasia, Type 1B and no experimental evidence demonstrating its impact on protein function have been reported. A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.238G>T, p.Val80Phe), supporting the critical relevance of codon 80 to EXOSC3 protein function. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
