This variant lies in the CYP17A1 gene (transcript NM_000102.4) at 3 bases into the intron immediately before coding-DNA position 1244, where C is replaced by A. Submitter rationale: Variant summary: CYP17A1 c.1244-3C>A alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Two predict the variant weakens a 3' acceptor site. Two predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.1244-3C>A has been observed in at least one individual affected with 17-hydroxylase deficiency (example: Zheng_2019). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
